Orlistat (English: Orlistat) is a weight loss drug with the trade name Xenical or Xenical. It requires a doctor’s prescription. However, in the United States, Orlistat is the first drug approved by the U.S. Food and Drug Administration. Approved weight loss pills that don't require a doctor's prescription.

Orlistat is a saturated derivative of lipstatin. Lipstatin is an effective natural pancreatic lipase inhibitor isolated from the bacterium Streptomyces toxytricini. It mainly acts on the gastrointestinal tract. It can inhibit the gastrointestinal tract required to digest fat. Enzymes, including pancreatic esters and gastric esters, reduce the absorption of fat by gastrointestinal esters to assist weight loss, but it still needs to be combined with exercise and diet to have the effect of losing weight.

Orlistat's effectiveness in promoting weight loss is well established. Data from clinical trials show that people given orlistat lost approximately 2-3 kilograms over a year compared to people who did not take the drug. In addition, Orlistat can moderately lower blood pressure. Some studies even point out that it can prevent the onset of type 2 diabetes, whether from weight loss itself or through other effects. In a large trial, orlistat was found to reduce the risk of diabetes by 40% in obese people with documented abnormalities in glucose metabolism.

Side effects of orlistat are mainly related to the gastrointestinal tract, including most commonly steatorrhea. However, its side effects will slow down with longer use. Orlistat is available without a prescription in Australia, the United States and the European Union. In the United States, over-the-counter approval is controversial, with consumer rights group Public Citizen repeatedly objecting on the grounds of safety and effectiveness. In Taiwan, it is currently the only legal auxiliary weight loss drug.

Orlistat is a drug used to treat obesity. In a one-year clinical trial, between 35.5% and 54.8% of subjects lost more than 5% of their weight, although not all of the weight lost was fat. And between 16.4% and 24.8% of subjects lost more than 10% of their weight. After stopping orlistat, a large number of subjects regained weight, regaining approximately 35% of the weight they had lost. Among obese people with type 2 diabetes, 6.2% of the total obese population remained obese after four years of taking orlistat, which was lower than the control group (9.0%) who took placebo [12] . Significant decreases in blood pressure were also seen in patients taking orlistat long-term (average decreases in systolic blood pressure of 2.5 mmHg and diastolic blood pressure of 1.9 mmHg).
