Duloxetine is a selective serotonin and norepinephrine reuptake inhibitor (SSNRI). The exact mechanism of duloxetine's antidepressant and central analgesic effects has not yet been clarified, but it is believed to be related to its enhancement of the serotonergic and norepinephrine functions of the central nervous system. Preclinical study results showed that duloxetine is a strong inhibitor of neuronal serotonin norepinephrine reuptake, and its inhibitory effect on dopamine reuptake is relatively weak. In vitro study results showed that duloxetine has no obvious affinity for dopamine receptors, adrenergic receptors, cholinergic receptors, histaminergic receptors, opioid receptors, glutamate receptors, and GABA receptors.
